El riesgo de estructuras proteicas laxas y "parcialmente desplegadas" como es el caso de la apolipoproteína A-I humana (apoA-I) es que pueden adoptar conformaciones anómalas que generan agregados insolubles. De hecho, fragmentos de apoA-I son encontrados en depósitos amiloideos de placas ateroscleróticas aórticas y en otros órganos. El procesamiento proteolítico pro-amiloidogénico parece ser favorecido por algunas mutaciones, como es el caso de Gly26Arg, pero ni los factores estructurales ni los ambientales que lo desencadenan son bien conocidos.
Con el objetivo de conocer los mecanismos moleculares que favorecen el plegamiento anómalo de la apoA-I se estudian, comparativamente con la proteína con la secuencia nativa, diferentes mutantes naturales que fueron identificadas en pacientes asociados a amiloidosis.
Se investigan los factores ambientales, en especial los relacionados a microambientes pro-inflamatorios (como estrés oxidativo) que favorecen tanto el procesamiento proteolítico como el plegamiento anómalo.
También es de nuestro interés conocer si formas mal plegadas de la proteína son a su vez capaces de activar la respuesta celular inflamatoria y de daño endotelial. Para tal fin disponemos de un modelo de respuesta inflamatoria que involucra células del sistema inmune y células endoteliales.
