Line overview

Our laboratory is dedicated to the study of the structure-function relationship of soluble Lipid-Binding Proteins (LBPs) since 1998, employing a range of biochemical, biophysical and molecular and cellular biology methods. We work in on in vitro systems based on recombinant proteins, model membranes and cell culture lines.

The original studies were focused on the comparative structure-function analysis of the Fatty Acid-Binding Proteins (FABPs) co-expressed in the intestinal epithelium: IFABP and LFABP. Biophysical and biochemical studies allowed us to describe the function of these proteins as fatty acid transporters for a better understanding of their participations in dietary lipids´ absorption and metabolism. We have demonstrated that, in spite of the similarities between these two isoformas in relation to tertiary structure and ligand binding characteristics, they employ very different ligand-transport mechanisms towards membranes. Furthermore, we have identified the structural domains of these proteins responsible of these differences, as well as of their different properties for interaction membrane of different composition.

Nowadays, our interest has been diversified towards the characterization of the specific functions of each FABP employing cell culture model systems. For example, we use a model system of intestinal epithelial cell, Caco-2 Cells, in which we diminish or augment the expression of the FABPs fos analyzing the impact of such alteration on the cellular proliferation and differentiation, on the metabolism of lipids, and the response to stress conditions and immflamatory processes. These studies are being extended to other systems, such as neuron cell model and cancerous cells. We are also developing pilot studies on patient´s samples to evaluate the diagnostic value of the levels of FABPs in serum as tissue injury biomarkers,e valuating in particular their use for differential diagnosis of gastrointestinal immflamatory pathologies that would help in the diagnosis of Celiac Disease.
